Kemik İliği Yağ Tabakasından Elde Edilen Mezenkimal Kök Hücrelerin Karakterizasyonu ve Subkutan Yağ ve Kemik İliği Kökenli Mezenkimal Kök Hücreleri ile Karşılaştırılması
Özet
Bone marrow adipose tissue (BMAT) has different properties than other adipose tissues. Recent studies have shown the effects of bone marrow adipose tissue on development / progression of acute myeloblastic leukemia (AML), and its contribution to bone metastases in different types of cancer. In this thesis study, characterization of mesenchymal stem cells (MSCs) obtained from fatty layer of bone (BMAT-MSC) of healthy donors and patients with Fanconi anemia (FA), who are known to be susceptible to leukemia was performed, their response to in-vitro adipogenic stimulation was assessed and compared with the results of bone marrow MSCs (BM-MSC) obtained from mononuclear cells by standard methods and with MSCs generated from subcutaneous fat tissue MSCs (SCF-MSC). In addition, the expressions of galectin-3, FABP4, Leptin-R, PDGFR-α, which are suggested to have a role in the interaction of leukemic cell with bone marrow microenvironment and in the pathogenesis of leukemia, were examined. MAT-MSCs, unlike other studies in the literature, obtained from the fatty layer of the BMAT-MSCs were shown to have differences in adipogenic gene expression profiles and response times to adipogenic stimulation when compared to those of BM-MSCs and SCF-MSCs. The adipogenic gene expression pattern of FA patients MSCs was also different compared to those from healthy donors suggesting that BM adipogenic dysregulation in FA patients. In co-culture experiments with AML (HL-60) cells, changes in surface molecule expressions were detected on MSCs. Further studies are required for confirmation of the preliminary results.
