Demir Oksit Nanoparçacıkları Üzerine Desteklenmiş Tabakalı Çift Hidroksitlere Anti-Kanser Ilaç Tutuklanması ve Tanecik Şekline Bağlı Salınım Davranışları
Özet
In this study, presented as a MSc thesis, nanocomposites for magnetic delivery of the
cancer drug 5- Fluorouracil (5FU) to the target tumor were prepared for the first time by supporting layered double hydroxides (LDHs) on magnetic core particles. The materials were characterized with a variety of chemical and physical techniques and drug release behaviors were examined. Chemical compositions of the products were determined by elemental analysis, structures were illuminated with PXRD, FTIR,
UV/Vis spectral techniques; magnetization, Zeta potential and BET surface area measurements. SEM-EDS and TEM were used for the morphological analysis of materials. Thermal stabilities were examined by TGA/DTA. Drug release profiles at
266 nm were obtained by UV-Vis spectrophotometer.
Nearly spherical magnetite and spindle-shaped hematite nanoparticles were used as the core materials. Mg-Al-LDH with interlayer nitrate ions was used as the Shell material. Bio-nanocomposities were prepared by exchanging interlayer nitrate ions
with the cancer drug 5-FU. TGA analysis results showed that the thermal stability of 5-FU was enhanced through intercalation into LDH layers. Effect of morphological properties on drug release was investigated and the observed complex drug release behavior indicated that more than one type of mechanism operates. Suitable kinetic models fitting the observed release profile were determined for each
sample by drawing the release versus time graphs. No significant change due to shape anisotropy was observed in in vitro release profiles however the
nanocomposites are expected to be more effective in in vivo conditions, in terms of shape-selective interactions with the target substrate.
