In this study, new hybrid monolithic structures were synthesized for use in affinity chromatography mode. This study was conducted to solve the previously encountered problems in the literature in which the increasing of cross-link density cause to prevent of the formation of mass transfer resistance regions. POSS-MA, with 10 methacrylate units, was utilized to overcome this problem as a cross-linker for synthesis of hybrid monolithic systems. For nano-liquid chromatography systems, MPBA-poly (POSS-MA) monolithic hybrid column was synthesized in one-step photopolymerization using thiol-ene chemistry method. Additionally, for borate affinity chromatography applications, VPBA-poly (POSS-MA) monolithic hybrid structure was synthesized in one-step method using free-radical polymerization in the presence of a thermal initiator. The surface morphology, specific surface areas (SSAs), and content of functional groups in the hybrid monolithic columns were determined using Scanning Electronic Microscope (SEM), nitrogen adsorption-desorption method and X-ray photoelectron spectroscopy (XPS), respectively.
The chromatographic performance of the MPBA-poly (POSS-MA) monolithic column (inner diameter: 100 μm, Length: 300 mm) was tested in the presence of alkylbenzenes, anilines and polycyclic aromatic hydrocarbons. It was found that the performance of column was independent from intended retention time. The VPBA-poly(POSS-MA) monolithic column was utilized in a microfluidic system using a silica capillary column (internal diameter: 300 μm, Length: 140 mm) for the isolation of diol containing biomolecules as well as the effect of loading concentration and flow rate on isolation yield was analysed. In this technique, the diol containing biomolecules (ß-NAD and ovalbumin) were isolated with satisfactory efficiency and good reproducibility as well.
