Rekombinant Fsh’nın Ivf Tedavisi Uygulanan Polikistik Over Sendromu Hastalarının Follikül Sıvılarından Elde Edilen Granüloza Hücrelerinde Konneksin 43 Üzerine Etkisi

The aim of this project is to investigate the effect of recombinant FSH (rFSH) on Connexin 43 (Cx43) activity of granulosa cells, that will be collected from the follicular fluids following isolation of the cumulus-oocyte complexes, with or without Anti Mullerian Hormone Receptor-II (AMHR2) antibody. After selecting oocyte-cumulus complex from the follicular fluid obtained during the OPU from 10 PCOS patients who referred to the IVF treatment for infertility, the granulosa cells were separated from the remaining follicular fluids and divided into 4 subgroups. After 2 hours culture in different combinations, Cx43 was evaluated by immunofluorescence staining in these groups. In this study, there were significant differences between the groups in terms of Cx43 staining rate and score. Significant differences were found between control and FSH group (P=0.011). We found significant differences between control and FSH+AMHR2 treated group (P=0.001). There was also significant difference between FSH and AMHR2 group (P=0.026). A significant difference was found between AMHR2 and FSH+AMHR2 treated group (P=0.002). Significant differences were found between control and FSH+AMHR2 treated group (P=0.001). A significant difference was found between AMHR2 and FSH+AMHR2 treated group (P=0.001). The result shows that rFSH affects the granulosa cells in PCOS patients and increases the expression of Cx43, thus enhancing the communication between the granulosa cells.
