Cancer is the one of the most difficult disease to detect and treat. By omic technologies it has been analyzed at molecular level about cancer. Among the omic technologies Proteomic, has been studied intensively worldwide, is a interdisciplinary area. Especially proteomics studies, which investigate about cancer at protein level, provide more information to support genomics results and clinic studies for cancer progression, detection and treatment. Liquid chromatography based proteomics researches have been making by using nano liquid chromatography. Although it gives more sensitivity, it has some drawbacks and is very difficult to control. In this thesis there are two man objectives. Firstly it has been tried to develop new ultra pressure liquid chromatography method to correct the drawbacks of nano liquid chromatography and to get reliable results. Bovine serum albumin was used as a standard protein to develop new analytical method and insight of the bovine serum albumin results CaCo2 colon cancer cell line was investigated. In second part of thesis effect of plat based hemostatic agent on colon cancer was investigated with new developed analytical method. Ultra pressure liquid chromatography was followed quadruple time of flight mass spectrometry. Mass spectroscopy results were evaluated with Maxquant. It was evaluated cancer targets and cancer pathways that are affected by hemostatic agent in colon cancer cells.
