In our study, the effects of N-acetylcysteine (NAC) and Calcium hydroxide (Ca(OH)2) on levels of Matrix metaolloproteinase-2, -9 (MMP-2, -9) and Tissue inhibitör metalloproteinase (TIMP-1, -2), which are released from lipopolysaccharide (LPS) stimulated human macrophage cell line, plays crucial role in apical periodontitis and chronic inflammation was evaluated comparatively. In our study, human monocyte precursor cells of the human acute monocyte leukemia cell lines (THP-1) was converted to macrophage cells. LPS-stimulated macrophage cells was incubated with NAC or Ca(OH). Following incubation; levels of tissue MMP-2, -9 and TIMP-1, -2 that controls the effectiveness of these group of enzymes was measured on protein and mRNA levels. As a result of this study NAC decreased the destruction enzymes in different time intervals.
