Eritropoetin Uygulamasının Mezenkimal Kök Hücre Gen ifade Profiline Etkisi
Abstract
Mesenchymal Stem Cells (MSC) have therapeutic importance because of their multipotency and immune modulation potentials. Erythropoietin is responsible for erytropoesis. However, EPO treatment of cells including mesenchymal stem cells, showed therapeutic effects. The aim of this thesis is to examine the effect of erythropoietin on bone marrow mesenchymal stem cells transcriptome and exosome derived miRNA profile. Firstly, effect of three different doses of EPO (1 IU/ml, 10 IU/ml ve 100 IU/ml) for 48 hours on MSC transcriptome profile was analyzed. The results illustrated that EPO treatment resulted in slight difference than control not exposed to EPO. The statistical analysis revealed that 10 IU/ml is the most different from the other groups. Therefore, small RNA libraries targeting miRNA was analyzed with 10 IU/ml EPO treated/not treated conditions with next generation sequencing. As a result, 44 miRNA showed different reading numbers. The expression of 22 was opened due to EPO treatment while the expression of 5 was closed because of same reason. Even though, 14 of miRNA illustrated decreasing reading number, only 2 of miRNA’s had increasing reading number. It will be important to perform studies in in vitro models mimicking different physiological conditions and diseases for MSC and exosome biology.
