The genus Gentiana has 400 species in the world and 12 species in Türkiye,
and plants belonging to this genus have had a wide range of uses in folk medicine for many
years. Gentiana olivieri Griseb. used in Southeastern and Eastern Anatolia as an appetite
stimulant, bitter tonic, and antidiabetic, as well as for the relief of stiffness and frostbite in
terrified people. In this region, it is known as "Afat" or "Afat otu". In this study, which is a
continuation of the studies carried out in our department, the aim was to determine the
phytochemical content of the aerial parts of Gentiana olivieri obtained commercially from the
Gaziantep region and to investigate their neuroprotective effects. For this purpose, decoction,
ethanol, water, chloroform and n-butanol extracts of G. olivieri were prepared, and their
neuroprotective effects were tested on the human neuroblastoma cell line SH-SY5Y. When the
effects on intracellular cytokine levels induced by LPS were evaluated, ethanol and water
extracts significantly increased IFN-α, aqueous extract IFN-β, chloroform and water extracts
c1q, n-butanol and chloroform c3 and aqueous extract c9 levels. 1 secoiridoide glycoside
(svertiamarin) and 1 oligosaccharide were purified from n-butanol extract by chromatographic
studies. The structures of the isolated compounds were elucidated using various spectroscopic
methods (13C NMR, 1H NMR, DEPT, and COSY techniques). In addition, the metabolites of
svertiamarin, sveroside, gentiopicroside and isoorientin, which were previously obtained from
Gentiana species, were detected in G. olivieri extracts by LC-qTOF-MS.
