Akut myeloblastik lösemi (aml) hastalığı hücrelerinin aptamer temelli akım sitometrik profillenmesi
Özet
AKCA O, Aptamer Based Flow Cytometric Profiling in Acute Myeloblastic Leukemia (AML) Disease Cells. Hacettepe University Graduate School of Health Sciences Stem Cell Program Master Thesis, Ankara, 2023. Aptamers are short, oligonucleotide sequences with a special 3-dimensional conformational structure and special binding sites. Aptamers can be produced specifically for the cell to be studied by the method called SELEX. Aptamers are seen as a molecule especially equivalent to antibodies compared to other affinity agents due to their low immunogenicity, more stable structure and ease of production.. In addition to various diagnostic methods, the diagnosis of AML is supported by the flow cytometry method, in which various surface proteins on the surface of the cell are stained with antibodies. In acute myeloid leukemia, there are differences in the surface proteins synthesized compared to healthy people. It has been shown in the literature that some proteins are overexpressed and some are less expressed. Aptamer binding studies were performed under the same conditions with cells from AML disease and cells from non-AML disease and flow stometric analyzes were performed. When compared with AML disease cells and non- AML cells, these different results still support studies that aptamers are equivalent to antibodies in the diagnosis of AML. Considering the attachment of aptamers to these surface proteins and the binding profiles they gave us in flow cytometry experiments, it will be possible to diagnose the disease through aptamers, and that the proteins to which aptamers bind are AML-specific proteins and that these proteins will be identified.
