MRI Kontrast Arttırıcı Ajan Olarak Magnetozom ve Hsa Kaplı Demiroksit Nanopartiküllerin Karşılaştırılması
Abstract
The efficiency of iron oxide nanoparticles, which were prepared by two different methods and used as MRI contrast enhancing agents, were identified. While one of the iron oxide nanoparticle was isolated from Magnetospirillum gryphiswaldense bacteria (called as magnetosome), the other one was prepared by double precipitation method and to enhance the biocompatibility of the nanoparticles, its surface was coated with human serum albumin protein. In the next step of the study, the morphological characteristics of the nanoparticles were determined by using the Scanning Transmission Electron Microscope (STEM), physicochemical properties by using the Fourier Transform Infrared Spectroscopy (FTIR) and the magnetic properties were determined by using of Electron Spin Resonance (ESR). In order to compare the magnetosome and iron oxide nanoparticle which were used as magnetic resonance contrast agents for imaging breast cancer, MDA-MB-231cell line were chosen as model for in vitro and vivo studies. In the study of cancer diagnosis and therapy, the difference between healthy and cancerous cells were exploited. The expression of EGFR (epidermal growth factor receptor) on the surface of the MDA-MB-231 cells is higher (about 90%) than the healthy cells.
