Transferrin Saflaştırılması İçin Manyetik İmmünoafinite Mikrokürelerin Sentezi

In this study, a new approach is developed for transferrin adsorption by immobilizing anti-transferrin antibody on monosize magnetic poli-glycidyl methacrylate microspheres with Fe3O4 nanopowder by dispersion polymerization. Synthesized micropspheres are characterized by FTIR, SEM, ESR, TGA-DTA, zeta sizer analysis. Swelling ratio of microspheres is investigated and calculated as %52. Microsphere density is calculated as 1.08 g cm3.Effect of concentration is investigated for determining the optimum immobilized anti-transferrin amount and is found as 2 mg g. Effect of pH, concentration and temperature is investigated for determining the optimum conditions for transferrin adsorption. Maximum adsorption capacity is found as 1.65 mg g at pH 6.0.Transferrin adsorption from artificial plasma is studied in magnetic environment and adsorption capacity is calculated as 0.12 mg g. In order to show that the desorption studies made no denaturation, SDS-PAGE studies are performed.
