Most of the biomaterials used nowadays are inspired by nature, so enhanced through biomimicing. Materials entegrated to biological systems should be provide morphological and chemical properties of them. Having porous structure is one of the most important property to increase proliferation of cells. Even there are several techniques to produce porous structures, most of them are technologically complicated and expensive. There is a technique, environmental friendly and not technologically complicated, called as Breath Figure (BF) method. Source of the most biomaterials explored by biomimicing is a polymer. Polyurethane (PU) firstly used in 60’s as biomaterial and it is one of the indespensable source of biomaterial in cardio vascular systems.
In the thesis, porous PU film was synthesized by BF approach to develop drug delivery system for cardio vascular system. As far known it is the first time porous PU film synthesized with the use of BF approach. As a result of characterization process the best polimer concentration is 5 mg.ml-1 to get the desired pore array. In addition, another essential parameter for the production of porous film by BF approach is relative humidity (RH) and the appropriate RH percentage depends on the type of polymer. Porous PU film with homegenous pore dispersion was best obtained at 75% RH and its morphological effects was examined at 5,10 and 20 min of humid atmosphere. The other parameter affects pore dispersion is time of humid atmosphere (exposure time) and the best value for production of porous PU film by BF approach was identified as 10 min. Morphological effect of exposure time examined at 55, 60, 65, 70, 75 and 80% RH respectively to determine the optimal curcimstance. As a result of morphological characterization the optimal curcimstances amoung the examined parameters were determined as 5 mg.ml-1 of polimer concentration, 75% RH of system atmosphere and 10 min of exposure time.
Compatibility of produced PU BF films to cardio vascular systems was tested through no toxic effects on human umblical vein cell line (HUVEC). Drug release experiments done with loading atorvastatin, one of the statins, to the films and gotten successful results. On the bases of results taken from the study PU films produced by BF approach is a promissing era to biomaterials.
