Okratoksin A Tayini İçin Nanolif Kaplı Kütle Hassas Immünosensör Hazırlanması
Abstract
Quartz crystal microbalance (QCM) surface was coated with cellulose acetate fiber via electrospinning method for increasing the surface area. It is expected to increase the sensitivity of mass sensitive immunosensor prepared for ochratoxin A (OTA) utilizing the high selective behavior of the antigen-antibody interaction. 1 mg/ml OTA antibody was immobilized on the blank QCM as control surface or electrospun coated QCM (ES-QCM) by means of chemically active aldehyde groups. The antigens that bound anti-OTA on ES-QCM were OTA (0.5-20 ng/ml) or BSA conjugated OTA (10-250 mg/ml). The amount of adsorbed OTA was determined by calculating the frequency shift due to binding of toxin on the surface. The calibration curves of the blank QCM and ES-QCM were obtained and these curves were compared with in terms of linearity and the detection limit. Selectivity of the immunosensor was tested with OTA like molecule citrinin (100, 250 ng/ml) and no cross-reactivity were observed.
