Mezenkimal Kök Hücrelerin Osteojenik Farklılaşmasının Bor Katkılı Hap-Kaplı Kitosan Doku İskelelerinde İncelenmesi
Özet
The aim of the present study is to investigate in-vitro osteogenic potential of mesenchymal stem cells, isolated from two different sources (bone marrow and adipose tissue), on boron doped hydroxyapatite (HAp) coated chitosan (B-HAp/chitosan) scaffolds with osteoinductive properties.In the first part of the study, chitosan scaffolds, which have approximately 100 μm pore diameter, were prepared by freeze-drying technique. In order to enhance the mechanical and bioactive properties of the scaffolds, they were coated with HAp and B-HAp through the aid of microwave energy by using 10xSBF (simulated body fluid) and boron containing 10xSBF (B-SBF). In the following part of the study, mesenchymal stem cells were isolated from adipose tissue and bone marrow. The cell viability was evaluated by Presto Blue analysis, cell morphology was investigated by F-actin/DAPI and crystal violet staining.
