Model Bir Yeşil Flöresan Protein İçin Aptamer Üretiminin ve Optik Biyosensörlerde Uygulanmasının Araştırılması
Abstract
The purpose of this research is identification of the target molecule-aptamer complex model that can be used as a model system to establish a measurement standard for increasing the efficiency of the analytic performance of the biosensor systems and Investigation the applicability of aptamer molecules. A fluorescence-based optical detection system was used as measurement device and the Green Fluorescent Protein (GFP) was used as fluorescent protein.
Initial RNA libraries, that had been developed to discover aptamer molecules against the target molecule (GFP) by performing SELEX procedure, were designed based on the previously selected aptamer molecules against the target molecule (GFP) with known primary sequence. Two distinct RNA libraries were synthesized as initial RNA libraries: (1) regional random oligonucleotide template and (2) fully random oligonucleotide template.
