Β-Estradiol Yüklü Doku İskeleleri ile In-Vitro Kemik Doku Mühendisliği
Abstract
The aim of the present study was to develop a system of 17-β estradiol (E2) loaded poly (lactic-co-glycolic acid) PLGA nanoparticles-chitosan/hydroxyapatite (HA) tissue scaffold and to investigate the in-vitro effectiveness of this system in adipose derived mesenchymal stem cell (AdMSC) cultures for bone tissue regeneration.
In the first part of the study, free and 17-β estradiol loaded PLGA nanoparticles were prepared from different compositions of PLA/PGA (50:50 and 65:35) by using emulsion-solvent evaporation technique. Characterization studies showed that free and 17-β estradiol loaded particle sizes were approximately 200 nm and 240 nm, respectively. Encapsulation efficiency of nanoparticles was calculated as 29% and 52% for 50:50 PLGA and 65:35 PLGA, respectively by using UV spectrophotometer and HPLC. Accordingly, 65:35 PLGA nanoparticles were selected as optimum carrier system. Release study from nanoparticles was continued up to 40 days and cumulative release (%) of 17-β estradiol from nanoparticles was determined as 45 %.
