Diklofenak Baskılanmış Yüzey Plazmon Rezonans Temelli Biyosensörün Hazırlanması

The aim of this study is the preparation of diclofenac (DFC) imprinted surface plasmon resonance sensor for detecting DFC. For this reason, SPR biosensors were modified with allyl mercaptane and the orientation of allyl groups was ensured. In the first step, the precomplex was prepared using DFC and methacrylic acid (MAA) monomer and prepared complex with crosslinker, EGDMA was polimerized on the golden surface of SPR under UV light. DFC imprinted SPR chip was characterized with FTIR-ATR, ellipsometry, atomic force microscopy (AFM) and contact angle measurements. A new chip was prepared without DFC in order to obtain DFC nonimprinted SPR sensor and the chip was characterized with ellipsometry, AFM and contact angle measurements. DFC sensing ability of DFC imprinted SPR sensor was investigated from DFC solutions with different concentrations. DFC solutions with different concentrations were used to determine the adsorption kinetics. Langmuir adsorption model was found as the most suitable model for DFC imprinted SPR chip. Competitive adsorptions of DFC and carbamazebine were investigated to show the selectivity of DFC imprinted SPR sensor DFC nonimprinted SPR sensor was prepared to determine imprinting selectivity of DFC imprinted SPR sensor. DFC imprinted SPR chip was investigated 10 times with same concentrations of DFC solution in order to show reuse of the chip.
