In this study, the production of phytase is aimed from a new microbial source that was isolated from natural sources. 12 different soil samples that were taken from barn and coop surroundings are used for the isolation of microorganism. K12 coded isolate among whole isolates is determined to be the highest producer of phytase.As a result of 16S rRNA sequencing analysis,K12 isolate was found Bacillus subtilis. The various production parameters of K12 isolate were optimized in order to ensure high enzyme production. In production medium, glucose was founded as the most suitable carbon source and ammonium nitrate (NH4NO3) was found to be the most suitable nitrogen source. In the presence of glucose, ammonium nitrate and sodium phytate and at the agitation speed of 150 rpm, 72 hours incubation at 30oC is said to be the most suitable conditions for the production of phytase.
