Momordıca Charantıa L. Ekstrelerinin İçeriğinin ve Melanoma Kanser Hücreleri Üzerine Etkisinin Metabolomik Çalışmalarla İncelenmesi

In this thesis study, Momordica charantia L. (bitter gourd) used in wound healing was investigated at the metabolome level with metabolomic studies based on Liquid Chromatography/Quadrapol Time-of-Flight Mass Spectrometry (LC/Q-TOF MS) and the in vitro effect of this plant on B16-F10 melanoma cell line was investigated by LC/Q-TOF MS. For this purpose, methanol extracts of the ripe fruit and seed of Momordica charantia L. were prepared. The extracts were analyzed using a gradient elution program with a C18 column using an LC/Q-TOF MS instrument. The raw data obtained were analyzed using various data
analysis methods. The detected peaks were normalized and the difference between the extracts of fruit and seed was examined. As a result of the research, it was seen that there was a difference in the metabolome level between fruit and seed. In addition, fruit and seed extracts were applied to B16F-10 melanoma cell lines to examine the efficacy of Momordica charantia L. in skin cancer. In order to observe the differences between the groups of control, fruit and seed, analysis was performed using LC/Q-TOF MS device. The differences between the control group and the results of the incubation of fruit and seed extracts in cell lines for 48 hours and 72 hours incubation were examined separately. As a result, it was observed that the metabolites of fruit (in cell) and seed (in cell) extracts were different from the control both for 48 hours incubation and after 72 hours incubation. It was concluded that this herb is promising for use in wound healing.
