dc.description.abstract | In dental surgery, various methods for reconstruction of bone defects and improving the insufficient amount of bone are available. Bone defects that cause trauma, infection, tumors, fractures and such cases, the use of autogenous grafts is gold standart for bone augmentation method; although there are still some disadvantages. Bisphosphonates are the most often preferred antiresorptive agents and used for treatment in postmenopausal osteoporosis, Paget's disease, bone metastases and such increased osteoclastic activity cases. Alendronate, the new generation bisphosphonate, reduces bone resorption in osteopenic individuals, increases the density, and increases bone formation due to anticatabolic feature. At the same time, it has also anabolic effects while strengthening the maturation and proliferation of osteoblasts. dc.description.ozet | Diş hekimliği cerrahisinde kemik defektlerinin rekonstrüksiyonu ve yetersiz kemik miktarının artırılması için çeşitli yöntemler mevcuttur. Travma, enfeksiyon, tümör, fraktür gibi kemik defektlerine neden olan durumlarda otojen greftlerin kullanımı ise; bazı dezavantajları olmasına rağmen hala kemik augmentasyon yöntemleri arasında altın standart olarak kabul edilmektedir. Bifosfonatlar; osteoklastik aktivitenin arttığı menapoz sonrası dönemde osteoporoz, Paget hastalığı, özellikle meme ve prostat kanserlerinde görülen kemik metastazı gibi durumlarda tedavi amaçlı kullanılan ve en sık tercih edilen antirezorptif ajanlardır. Osteopenik bireylerde kemik rezorpsiyonunu azaltarak dansitesini artıran yeni nesil bifosfonatlardan alendronat, antikatabolik özelliğinden dolayı osteoklastik aktiviteyi inhibe ederek kemik formasyonunu artırmaktadır. Aynı zamanda osteoblastların proliferasyonu ve maturasyonunu da güçlendirerek anabolik etkiye de sahiptir. Bu çalışmada intramembranöz kemikleşme modeli ile kemikleşen çene kemiklerinden elde edilen otojen greftlerin başarısını artırmak amaçlı lokal alendronat kullanımının etkinliğinin tavşan defekt modeli üzerinde histomorfometrik ve radyolojik olarak incelenmesi amaçlanmaktadır. | tr_TR |
