The aim of this study is to investigate the presence, number and proliferation capacity of mesenchymal stem cells in the dental pulp of young and old individuals with healthy, reversible pulpitis and irreversible pulpitis. In this study, healthy pulps were obtained from extracted wisdom teeth and inflamed pulps were obtained during root canal treatment. Study groups as follow; Young healthy pulp tissues(n=2), young pulp tissues with reversibl pulpitis (n=2), young pulp tissues with irreversibl pulpitis(n=2), old healthy pulp tissues(n=1), old pulp tissues with irreversibl pulpitis(n=1). Primer cell culture were set up witk obtained pulp. Mesenchymal stem cell specificity of proliferating fibroblast cells was examined by flow cytometry method and immunofluorescence staining method. The proliferation potential of the obtained stem cells was analyzed by lateral proliferation method. Stem cells from old healthy pulp proliferate more slowly than those from young healthy pulp, while young reversible pulpitis stem cells proliferated faster than young healthy pulp stem cells. According to the immunhistochemical marking method, it was observed that CD73, CD90 and CD105 antibodies were commonly labeled as positive mesenchymal stem cell surface markers in the cell stoplasmas of all samples, and CD34 and CD45 were not marked as negative surface markers. According to the flow cytometry results, the mesenchymal stem cell specificity of all groups was found between 98% and 100%, without any difference between the groups. As a result, while mesenchymal stem cell specificity of the dental pulp did not change due to inflammation and age, age-related decrease in proliferation capacity was observed.
