Cancer is a group of diseases that ranks second in the list of causes of death and therefore early diagnosis and treatment are of vital importance. Liquid biopsy, which is a promising method in the clinic, makes it possible to profile the cancer and following its prognosis, if effective detection, capture and separation of circulating tumor cells (CTCs) can be performed on a simple blood sample taken from the patient. Cause of this goal, many different principles of CTC separation techniques have been tried and technologies have been developed. In this thesis study, with the aim of catching and separating CTCs that leave the primary tumor region, carried in the blood stream and cause metastasis: An affinity system based on poly(2-hydroxyethyl methacrylate) PHEMA cryogel containing Concanavalin A (Con A) lectin as ligand has been developed. The ability of Con A lectin, MCF-7 and MDA-MB-231 breast cancer cells to effectively recognize and bind thanks to their changing surface molecules and increasing sugar structure, unlike normal cells, is the basis of the affinity chromatography in this study. In order for the affinity system to work effectively, the selection of the ideal cryogel material is great importance. For this purpose: Cryogel groups with different properties were created depending on variables such as varying bovine serum albumin (BSA) concentrations in its content and whether it contains glutaraldehyde, which is the crosslinker of BSA. The effects of these variables on cryogels; Comparison of the success of cryogel groups with each other in the light of data obtained as a result of various characterization analyzes (Swelling experiments, SEM imaging, Micro-CT and FTIR) and in-vitro cell experiments (cell counting with Trypan blue, MTT and SEM imaging in cellular samples). Evaluated through at the end of all experimental and analysis studies, the most successful cryogel group, so the ideal material is: PHEMA-G-BSA100-ConA with Con A bound, containing the highest concentration of BSA and its crosslinker glutaraldehyde in its structure. (Swelling rate of 681.4%, macropore amount of 77.6%, binding of 4.478 mg Con A lectin per 1 gram polymer, and having the highest average % viability in in-vitro experiments in incubation times). This developed system has the potential to be used in various diagnostic and therapeutic studies such as cancer profiling with these cancer groups.
