Kanser Hücrelerinin Aptamer ve Antibadi Taşıyan Manyetik Nanopartiküller ile Hedeflenmesi
Özet
Within the context of this thesis, developing of an early diagnosis nanoparticular system in order to detect and image human breast cancer cell line of MDA MB 231 is aimed. In this regard, gold coated superparamagnetic nanoparticles (SPIONs) were designed and characterized. Average sizes of synthesized SPIONs and gold-coated SPIONs which were determined by TEM were detected as 5.62 + 1.49 nm and 57.04 + 8.58 nm, respectively. FITC-labeled aptamer and antibody molecules were immobilized onto gold-coated SPIONs and than developed carrier system was applied to MDA MB 231 breast cancer cells in the cell culture medium. Flow cytometry was used to determine the interaction rates between tumor cell receptors and fluorescently labeled aptamer and antibody molecules and antibody-immobilized SPION in the concentration of 108 µg mL have had the highest conjugation rate by the 64.4%. Tumor cell selectivity was also imaged by the technic of Fluorescent Microscopy.
