In this study, we aimed to obtain an artificial bilayer skin substitute by combining human dermal fibroblasts (Detroit 551) cultured on chitosan-gelatin superporous hydrogels (SPHs) as a dermal layer and human keratinocytes (HS2) suspended in fibrin gel as an epidermal layer.A novel chitosan-gelatin scaffold was prepared in superporous hydrogel (SPH) form by microwave irradiation-supported gas foaming method. Sodium bicarbonate was used as gas blowing agent and crosslinking was achieved by means of glyoxal. Average pore diameters of the scaffolds were calculated from SEM (Scanning Electron Microscope) photographs as 509 101 μm and elastic modulus determined by compression test was 0.01 MPa. The water uptake capacity, swelling ratio and time to reach equilibrium for chitosan-gelatin SPHs were found to be 48%, 42.42 2.94, and 60 sec, respectively. Void fraction and density of the SPHs were calculated as 0.22 0.08 mL g, 0.07 0.00 g mL, respectively.
