Sesamum İndicum L. (Susam) X Papaver Somniferum L. (Haşhaş) Arasındaki Çapraz Reaksiyonların Western Blot Yöntemi ile Gösterilmesi

The prevalence of food allergies is increasing worldwide for uncertain reasons. lgE mediated food allergy affects approximately 3% of the population. While sesame allergy is among the most commonly diagnosed food allergies, the incidence of poppy allergy is low, but it is known to cause serious reactions. Since researchers began to characterize the molecular properties of food allergens, they have learned about the cross-reactivity of different foods. More importantly, this has been useful in predicting the severity and types of allergic reactions in patients. Within the scope of the thesis, it is aimed to show the cross-reaction status as a result of Western Blot and inhibition experiments after categorizing the patients according to their ability to consume sesame and / or poppy in 20 patients who underwent provocation test according to SPT and specific IgE results. In conclusion, we identified previously defined bands of approximately 9 kDa, 15 kDa, 17 kDa, 45 kDa, 52 kDa, 57 kDa in patients in our clinic, which are thought to belong to sesame by size. We identified previously undefined bands in the range of 65-80 kDa, 80-115 kDa range, approximately 9 kDa, 22 kDa, 25 kDa, 28 kDa, 30 kDa, 35 kDa, 40 kDa, 50 kDa, 65 kDa thought to belong to sesame. Previously identified bands in the molecular weight of 14 kDa, 17 kDa, 20 kDa, 25 kDa, 30 kDa, 40 kDa and 45 kDa which are thought to belong to poppy by size, unidentified bands in the molecular weight of 9 kDa and bands in the range of 15-25 kDa, 25-30 kDa, 30-40 kDa, 50-65 kDa are shown. From these bands approximately 15 kDa, 17 kDa, 25 kDa, 30 kDa, 40 kDa, 45 kDa, 52 kDa, 57 kDa, 65 kDa molecular weight, 10-15 kDa, 15-25 kDa, 25-30 kDa, 30-40 kDa, 65-80 kDa range it was determined that sesame bands were inhibited by the poppy allergen. Poppy (Papaver somniferum L.) bands in the range of 14 kDa, 17 kDa, 45 kDa, 25-30 kDa, 30-40 kDa, 80-115 kDa were also partly inhibited by the sesame allergen. As a result of Western analysis and inhibition experiments, there is a cross-reaction between sesame (Sesamum indicum L.) and poppy (Papaver somniferum L.) foods. We believe that the demonstration of the cross-reaction will be useful in reducing the need for food provocation testing by predicting the reaction severity, determining the responsible allergen and making clinical diagnosis, and separating the true sensitization from the sensitization that develops due to cross-reaction.
