Mezenşimal Kök Hücrelerden Farklılaştırılmış İşaretli Kardiyomiyositlerin In Vivo Implantasyon Sonrası Ektopik Göç Potansiyelinin İncelenmesi
Özet
In this study, mesenchymal stem cells were isolated from the rat fat tissue. Obtained cells were labeled by BrdU after mesenchymal stem cells were differentiated by 5-Aza to cardiomyocytes. Acellularised cardiac tissue scaffolds were acquired by applying of ionic and non ionic detergents and labeled differentiated cells were implemented to the obtained asellularized scaffolds. Implantation of these cells structure material composite to the rat retroperitoneum and ectopic migration of the implanted cells were examined by different techniques.The rat gonadal and subcutaneous adipose tissue was isolated in the first part of the study. 5-Aza, a chemical agent, was used as a cardiomyogenic differentiation factor. The medium was changed twice per week during the differentiation procedure and the cells were incubated for four weeks at 37 °C and 5%CO2- 95%O2 in standard culture conditions.
