Periodontal diseases are considered one of the chronic complication of Type 2 diabetes. Both diabetes and periodontal diseases are complex, multifactorial diseases to which genetic factors are thought to predispose individuals. Hypoxia inducible factor -1α (HIF-1α) is a transcriptional activator of vascular endotelial growth factor (VEGF) and is critical for angiogenic responses to hypoxia. Tumor necrosis factor-α, which is a major proinflammatory cytokine, activate the HIF-1α pathway in various cells. The TNF-α /HIF-1 /VEGF pathway might have a regulatory function in periodontal tissues. Our aim was to investigate potential effects of HIF-1α +1772C>T, VEGF +405G>C, TNF-α 308G>A single nucleotide polymorphisms (SNP) on periodontal diseases in Type 2 diabetics. A total 116 individuals were enrolled in the study including 24 chronic periodontitis with Type 2 DM, 35 gingivitis with Type 2 DM, 7 healthy periodontal status with Type 2 DM, 26 chronic periodontitis and 24 healthy periodontal status. Allelic and genotypic comparisons between healthy periodontal status and other groups were evaluated by PCR-RFLP. We found a significant association between the A allel at TNF-α 308G>A and risk of gingivitis on Type 2 DM (OR=3.75, CI: 1.015 – 13.860, P=0.048). There was no significant association detected between HIF-1α 1772C> T, VEGF 405G>C polymorphisms and risk for periodontal diseases with Type 2 DM. Future studies may contribute to the investigation of the potential polygenic predisposition of the diseases and reinforce our findings.
