dc.description.abstract | Perfluorooctanoic acid (PFOA)
is a persistent environmental pollutant of toxicological importance. However,
its mechanism of toxicity was not well-characterized. The aim of this study was
to investigate the possible cytotoxic and oxidative stress inducing effects of
PFOA in in vivo and in vitro model systems in the presence or absence of
taurine and coenzyme Q10 (CoQ10). For this purpose, in the in vivo part of the
study, animals were treated with two different concentrations (15 or 30
mg/kg/day) of PFOA by oral route for 10 days and lipid peroxidation, total
glutathione levels, and activities of antioxidant enzymes (catalase, glutathione
peroxidase and Cu-Zn-superoxide dismutase) were measured in liver, brain
and kidney tissues. A significant and dose-dependent decrease in body
weights and hepatomegaly in addition to reduction in kidney weights were
observed in animals treated with PFOA. Evaluation of MDA levels and
enzymatic and non-enzymatic antioxidant parameters showed that especially
lower concentration of PFOA induced lipid peroxidation and a modification on
the antioxidant systems. Pre-treatment of mice with taurine or CoQ10 before
the administration of 30 mg/kg of PFOA provided a marked protection against
PFOA-induced lipid peroxidation in liver tissue. In the in vitro part of the study,
cytotoxic effects of PFOA was determined by MTT assay, and it was found that
the effect of the substance on the viability of vero cells was time dependent.
Cytotoxicity was observed ~≥ 200 μM concentrations of PFOA. Taurine or
CoQ10 pretreatment was partially protective against PFOA-induced
cytotoxicity. These results suggest the involvement of reactive oxygen species

toksikolojik öneme sahip kalıcı bir çevresel kirleticidir. Ancak toksik etki
mekanizması tam olarak tanımlanmamıştır. Bu çalışmada, PFOA’nın olası
sitotoksik ve oksidatif stres indükleyici etkileri taurin ve koenzim Q10 (KoQ10)
varlığı ve yokluğunda in vivo ve in vitro model sistemlerde araştırılmıştır. Bu
amaçla, çalışmanın in vivo kısmında hayvanlar, PFOA’nın iki farklı
konsantrasyonuna (15 veya 30 mg/kg/gün) oral yoldan 10 gün süreyle maruz
bırakılmışlar ve karaciğer, beyin ve böbrek dokularında, lipid peroksidasyon,
toplam glutatyon düzeyleri ve antioksidan enzim aktiviteleri (katalaz, glutatyon
peroksidaz ve Cu-Zn süperoksit dismutaz) ölçülmüştür. PFOA uygulanan
hayvanlarda vücut ağırlıklarında önemli ve doza bağımlı bir azalma ile böbrek
ağırlıklarında azalmaya ek olarak hepatomegali gözlenmiştir. MDA
düzeylerinin ve enzimatik ve enzimatik olmayan antioksidan parametrelerin
değerlendirilmesi, özellikle daha düşük konsantrasyonda PFOA’nın lipid
peroksidasyonu ve antioksidan sistemlerde değişikliği indüklediğini
göstermiştir. Farelerin 30 mg/kg PFOA verilmeden önce taurin veya KoQ10’a
ön-maruziyeti karaciğer dokusunda PFOA’nın indüklediği lipid peroksidasyona
karşı belirgin bir koruma sağlamıştır. Çalışmanın in vitro bölümünde PFOA’nın
sitotoksik etkileri MTT deneyi ile belirlenmiş ve bileşiğin vero hücrelerinin
canlılığı üzerindeki etkisinin zamana bağımlı olduğu bulunmuştur.
Sitotoksisite, PFOA’nın ~≥ 200 μM konsantrasyonlarında gözlenmiştir. Taurin
ve KoQ10 ön uygulamasının PFOA’nın indüklediği sitotoksisiteye karşı kısmen
koruyucu olduğu saptanmıştır. Bu sonuçlar, PFOA’nın toksisite
mekanizmasında reaktif oksijen türlerinin yer aldığına işaret etmektedir. | tr_TR |
