Synthesıs of Condensed 1,4-Dıhydropyrıdıne Derıvatıves and Evaluatıon of Theır Calcıum Channel Modulatıng and Antıoxıdant Actıvıtıes

In this study, study, a pharmacophore model based on calcium channel blocker ligands was generated by LigandScout and nineteen 2-(methacryloyloxy)ethyl 4-aryl-2,6,6-trimethyl-5-oxo-1,4,5,6,7,8-hexahydroquinoline-3-carboxylate derivatives were synthesized via multicomponent one pot Hantzsch
reaction of 4,4-dimethylcyclohexane-1,3-dione, 2-(methacryloyloxy)ethyl
acetoacetate, substituted benzaldehyde and ammonium acetate in methanol under
microwave irradiation in a short time. The structures of the synthesized compounds
were identified by IR, 1H-NMR, 13C-NMR and mass spectroscopy; and were proved
by elemental analysis. The calcium modulating activity of the compounds were
determined by testing their myorelaxant effect on isolated strips of rabbit gastric
fundus smooth muscle by using nifedipine as standard and the antioxidant activity was
determined by applying differential pulse voltammetry technique. The results showed
the compounds exerted concentration-dependent relaxation responses on gastric fundus
smooth muscle strips but the compounds were less potent than nifedipine; and some of
the compounds exhibited good antioxidant activity.
