dc.description.abstractThe genus Gentiana (Gentianaceae) consists species with important phytochemical properties and have been widely used in traditional folk medicine. Iridoids, secoiridoids, C-glycosylflavones and xanthones are considered as the most promising groups of compounds responsible for the pharmacological activities of the species. Moreover, they are also evaluated as the important key compounds in the aspect of chemotaxonomy of the genus Gentiana. The aim of this thesis, which was planned as a continuation of our previous studies on Gentiana species in our department, was to identify the chemical composition as well as MAO-A enzyme inhibition activities of the overground parts of Gentiana pyrenaica, G. verna subsp. balcanica and G. verna subsp. pontica, growing on Uludağ (Bursa) and Zigana Mts. (Trabzon). n-buthanol extracts of the over ground parts of the title plants were prepared and the MAO-A inhibitory activity of these extracts were tested. By means of chromatographic studies on the H2O extracts a new secoiridoid glucoside, three new benzophenone glucosides in addition to two xanthone glucosides were isolated from G. verna subsp. pontica; and a flavon Cglucoside and two simple phenolic glucosides were isolated from G.pyrenaica. Chemical composition of G. verna subsp. balcanica were analyzed by using an HPLC method. Gentiana pyrenaica, G. verna subsp. balcanica and G. verna subsp. pontica and two benzophenone glucosides were found to possess MAO-A inhibitory activity.tr_TR

dc.description.ozetGentiana (Gentianaceae) cinsi, bitki kimyası açısından ilginç özellikler gösteren ve uzun yıllardan beri geleneksel halk tıbbında geniş bir şekilde kullanılmakta olan türlere sahiptir. Gentiana türlerinin kimyasal yapısında yer alan iridoit, sekoiridoit, flavonoit C-glikoziti ve ksantonlar, bu türlerin gösterdiği farmakolojik etkilerden sorumlu bileşikler olarak kabul edilirler. Öte yandan, aynı bileşikler Gentiana cinsinin kemotaksonomisi açısından da önemli birer anahtar olarak değerlendirilirler. Önceki yıllarda, Anabilim Dalımızda Gentiana türleri üzerinde gerçekleştirilen çalışmaların devamı niteliğinde planlanan bu tez çalışmasında, Uludağ (Bursa) ve Ziganalar (Trabzon)’da yetişmekte olan Gentiana pyrenaica, G. verna subsp. balcanica ve G. verna subsp. pontica türlerinin toprak üstü kısımlarının kimyasal yapısının aydınlatılması ve MAO-A enzim inhibisyonu etkilerinin araştırılması hedeflenmiştir. Bu amaçla Gentiana pyrenaica, G. verna subsp. balcanica ve G. verna subsp. pontica türlerinin toprak üstü kısımlarının nbutanollü ekstreleri hazırlanmış, bu ekstrelerin MAO-A inhibisyonu etkisi test edilmiştir. Sulu ekstreler üzerinde gerçekleştirilen kromatografik çalışmalar sonunda G. verna subsp. pontica’dan 1 yeni sekoiridoit glukoziti, 3 yeni benzofenon glukoziti yanı sıra 2 ksanton glukoziti; G. pyrenaica’dan 1 flavon C-glukoziti ve 2 basit fenol glukoziti izole edilmiştir. G. verna subsp. balcanica’nın kimyasal yapısı YBSK sistemi kullanılarak araştırılmıştır. Gentiana pyrenaica, G. verna subsp. balcanica ve G. verna subsp. pontica ekstrelerinin ve iki benzofenon glukozitinin MAO-A enzim inhbisyonu yaptıkları belirlenmiştir.tr_TR
